ultrastructural studies of osteoblast cells in osteopetrotic op/op mouse

چکیده

introduction: the osteopetrosis is a common disease in man and animals which is characterized by calcium accumulation in bones. the purpose of this study is to eveluate the osteoblast ultrastructure in osteoptrotic op/op mouse. material and methods: in this study 5 op/op (experimental) and 5 control mouse were killed by overdose of choloroform. their femur bones were taken, decalcified by edta, fiexed in glutaraldehyde and osmium tetroxide, embeded in epon, sectioned by diamond knife, stained by lead citrate and uranil acetate and photographed by transmission electron microscope (hitachi 800, japan). results: the ultrastructural observation of osteoblast cells revealed a decrease in the mitochonderia and endoplasmic reticulum in of op/op mouse, but the cytoplasmic vacuoles and free ribosomes were high. the nuclear envelope in op/op group was irregular and higher chromatin density. also, the osteoid in op/op group was thickened. conclusion: the activity of osteoblast was decreased due to the inactivity of osteoclast as a result of lack macrophage colony stimulating factor.

Ultrastructural studies of the mouse blastocyst substages.

The mouse blastocyst stage covers approximately 2 days. During this period, embryonic development advances through four ultrastructurally distinct substages. Specific ultrastructural characteristics, such as changes in cell shape and/or the distribution of intracellular organelles, may be used to characterize the various cell types from each other at each substage (e.g. trophoblast from inner c...

Morphologic Variation in the Osteopetrotic Op/Op Mouse Skeleton

SUMMARY This study is designed to confirm the changes of various bones in the osteopetrotic op/op mouse. The female adult mouse were sonicated in papain solution. After the complete treatment, the bones (Skull, Mandible, Scapula, Humerus, Radius, Ulna, Hip, Femure, Tibia, Atlas, Axis and 5th Lumbar vertebra) were measured three dimensionally using of high quality scale and low power binocular ...

Ultrastructural and quantitative studies of mammary tumor virus production in cultured mouse mammary tumor cells.

A whole-cell-mounting technique was successfully applied to the study of mouse mammary tumor virus budding from cells in tissue culture. The technique permitted the visualiza tion of ultrastructural detail, including the cores and the surface spikes of budding virions. The latter are difficult to discriminate in thin sections. Budding mammary tumor virus was compared with budding murine leukemi...

Megaoesophagus in the mouse: histochemical and ultrastructural studies.

Histochemical and ultrastructural studies of the muscle coat of the oesophagus from ICRC/HiCri mice (with megaoesophagus) and DBA/2fNCri mice (normal oesophagus) were carried out. The striking observation from histochemical studies was the presence of smooth muscle in the abdominal segment of the oesophagus from ICRC mouse in contrast to the control strain where smooth muscle was present only i...

Identification of macrophages and dendritic cells in the osteopetrotic (op/op) mouse.

We used a panel of monoclonal antibodies and immunocytochemistry to identify macrophages and dendritic cells in mice that are deficient in macrophage colony stimulating factor (M-CSF or CSF-1) because of the recessive osteopetrotic (op/op) mutation. Prior work had shown that osteopetrosis is associated with a lack of osteoclasts, phagocytic cells required for remodelling in bone. Additional mac...
